Design: Whole genomic DNA of was digested with PstI and MspI and ligated to adapters such that the barcode (GATCGCATTT for this sample) was adjacent to the PstI cut site. After pooling of 95 barcoded samples, size selection from 200-500 bp was performed on a 2% agarose gel. The library was then amplified with KAPA Hi-Fi polymerase. Sequencing was performed beginning at the barcoded PstI end of fragments. Samples were demultiplexed by barcode, and adapter and chimeric sequence was trimmed, using TagDigger 1.0.
